Purchase of a multiphoton microscope instrument package

Aarhus Universitet

With the multiphoton microscope with galvo scanning system (Item A), we will perform calcium imaging of retinal neurons and also perform two photon targeted patch-clamp recordings of fluorescently-labelled retinal neurons.
In order to perform such experiments, the multiphoton microscope must allow simultaneous two-photon imaging and infrared (around 750 nm) camera imaging. We will use DLP overhead projector for both a light source for infrared imaging and visual stimulation.
This could be achieved by moving sample stage and manipulaters as one in X-Y dimension, in relative to fixed microscope in order not to disturb the light path for visual stimulation.
We will also perform calcium imaging from brain of head-fixed mice with the multiphoton microscope with galvo scanning system.
The microscope must be equipped with two PMTs for imaging green and red, and one should be high-sensitive GaAsP PMT to detect signal from deep brain.
With the multiphoton microscope with resonant scanning system (Item B), we will perform deep brain calcium imaging from head-fixed behaving mice.
In order to perform such experiments, the scanning must be very fast (30 frames per second with 512 x 512 pixels) and should have large space for sample. Objective must be tiltable to allow imaging from side of brain.
The microscope must be equipped with two PMTs for imaging green and red, and one should be high-sensitive GaAsP PMT to detect faint signal from deep brain.
The software offered must be user-friendly and control subparts (pockel cell). The imaging must be able to be triggered by TTL.
Optical parts offered should split the laser into the two microscopes and direct the lasers into scanning path for the two microscopes.
Due to the restriction of the size of experimental room, 2 multiphoton microscopes and one laser must be installed on one optical table with the size of 3 m x 1.5 m, which require compact foot print and small dimension.
The two multiphoton microscopes offered must be installed after the optical table and laser is installed in the experimental room, cf. Appendix 2 — Specification of requirements.
Therefore the flexibility related to delivery and installation of the 2 multiphoton microscopes will be a part of the procurement procedure.
The Contracting Authority puts out to tender the purchase of the said microscope instrument package and includes issues concerning delivery, installation, training, guarantee and service contract as a part of the contract.
