Purchase of microscope
The Contracting Authority intends to purchase an instrument based on fluorescence microscopy for in vivo imaging of blood flow in single capillaries and simultaneous measurement of morphological and biochemical parameters of endothelial cells, pericytes and associated tissues (the Microscope).
The aim of the instrument is to enable the simultaneous measurement of red blood cell flux in as many individual brain capillaries as possible within the field of view, and at the same time record morphological changes in cells constituting the capillary wall and biochemical changes in surrounding parenchyma in order to study the interplay of pericytes, capillary bloodflow and tissue activity over time. For this purpose two-photon microscopy is chosen as the main methodology, optionally supplemented by laser scanning confocal microscopy. The instrument should be able perform in vivo imaging in anaesthetized rats and mice, which may at the same time be subject to artificial ventilation, iv infusion, electrophysiological measurements.
The instrument will be established as a basic unit, which should be able to operate as a stand-alone system for two-photon in vivo imaging of rats and mice.
